The 4SM16 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8 binds to MHC class I and, through its association with protein tyrosine kinase p56lck, plays a key role in T cell development and activation.

This antibody is recommended for immunohistochemistry of formalin-fixed paraffin-embedded (FFPE) or frozen mouse tissue and can also be used for staining mouse neural tissue.

Microscopy and immunohistochemical staining of frozen and FFPE tissue sections.

Applications Tested:

• Tested by IHC on FFPE mouse tissue using low/high pH antigen retrieval ≤ 5 µg/mL.
• Tested by IHC on frozen mouse tissue ≤ 5 µg/mL.
• Optimal performance requires titration per assay.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein involved in signal transduction, T cell differentiation, and activation. CD8 primarily exists as a CD8αβ heterodimer acting as a co-receptor with TCR for MHC class I molecules. CD8 is also found as CD8αα homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes recruits Lck kinase, inducing lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance, with differentiation enhanced by IL-2, IL-12, and TGF-β1.
