Description:
The AISB12 monoclonal antibody reacts with mouse CD20, a B cell-specific 32 kDa membrane protein involved in intracellular calcium regulation. CD20 expression begins after CD19 induction during development and increases during maturation, with most mature B cells expressing CD20. The human CD20 antigen is a well-established immunotherapeutic target in B cell lymphoma and several autoimmune diseases such as rheumatoid arthritis and type I diabetes. CD20 has four transmembrane domains and a 44 amino acid extracellular loop domain where AISB12 binds.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, as well as a subset of T lymphocytes and follicular dendritic cells. It regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation. CD20 is associated with lipid rafts, with association intensity depending on extracellular triggers and BCR aggregation. After receptor ligation, CD20 remains at the cell surface while BCR undergoes endocytosis. Because CD20 is highly expressed on most B-cell malignancies and remains membrane-bound after antibody treatment, it serves as an effective target for antibody-mediated B cell depletion.

Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
