Product Specific Information

Description:
The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32.
CD16 (Fc gamma III Receptor) and CD32 (Fc gamma II Receptor) are low affinity receptors for the mouse IgG Fc portion and are expressed by B cells, monocyte/macrophages, NK cells, and neutrophils.

Applications Reported:

• Reported for use in flow cytometric analysis and Fc receptor blocking in functional studies.
• For blocking Fc receptors in flow cytometric analysis, pre-incubate cells with 0.5–1 µg of purified anti-CD16/CD32 per million cells for 5–10 minutes on ice prior to staining.
• For functional assays, use Functional Grade purified 93 (Product #16-0161).

Applications Tested:

• Tested by flow cytometric analysis of mouse splenocytes.
• Recommended ≤0.5 µg per test (100 µL final volume).
• Optimal titration recommended depending on cell number (10⁵–10⁸ cells/test).

Target Information

CD16 is a 50–65 kDa cell surface molecule expressed in two forms: a transmembrane form on NK cells and some T cells, and a phosphatidylinositol-linked form on granulocytes.
It acts as a low-affinity receptor for IgG (FcR III) and mediates antibody-dependent cell-mediated cytotoxicity (ADCC).

CD32 is a 40 kDa glycoprotein functioning as a low-affinity receptor for IgG (FcγRII), mediating endocytosis, secretion activation, cytotoxicity, and immunomodulation.
It is expressed by B cells, monocytes, granulocytes, and platelets, and involved in immune complex phagocytosis and antibody production regulation.
Certain CD32 gene variants are associated with increased susceptibility to systemic lupus erythematosus (SLE).
Multiple transcript variants encoding different isoforms of CD32 have been identified.
