Description:
The 8D4-8 antibody reacts with human interleukin-4 (IL-4), a 15–19 kDa cytokine secreted by Th2 cells.

Applications Reported:
Used as a capture antibody for human IL-4 ELISA and for intracellular staining in flow cytometric analysis.

Applications Tested:
Validated by intracellular flow cytometric analysis of stimulated normal human peripheral blood cells. Recommended usage ≤ 0.25 µg/test (final volume 100 µL). Cell number may range from 10⁵ to 10⁸ cells/test. Optimize titration for best performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells and serves as a ligand for the IL-4 receptor. It shares overlapping functions with IL-13 via receptor binding. STAT6 mediates IL-4 signaling and immune regulation. The IL-4 gene is part of a cytokine cluster (IL-3, IL-5, IL-13, CSF2) on chromosome 5q, closely located near IL-13 and regulated by long-range elements. IL-4 delta 2 is a known splice variant.
