Product Specific Information

The 6.7 monoclonal antibody reacts with human and non-human primate (rhesus or cynomologus) CD18, also known as beta2 integrin. CD18 is a 90–95 kDa type I transmembrane protein that heterodimerizes with CD11a, CD11b, and CD11c. The CD18/CD11a complex, known as LFA-1, plays an important role in cellular adhesion and inflammatory reactions.

CD18 is broadly expressed by human lymphocytes and monocytes. Peripheral blood granulocytes also express CD18 at lower levels. The 6.7 monoclonal antibody can induce proliferation of resting T cells cultured with monocytes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human blood cells. Use at 5 µL (0.8 µg) per test (final volume 100 µL). Cell number should range from 10⁵ to 10⁸ cells/test.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample (10³–10⁸ cells).
• For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD18 integrin beta 2 subunit is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11a–d molecules to constitute leukocyte (beta2) integrins:

• αLβ2 (CD11a/CD18, LFA-1)
• αMβ2 (CD11b/CD18, Mac-1, CR3)
• αXβ2 (CD11c/CD18)
• αDβ2 (CD11d/CD18)

These integrins are essential for leukocyte migration and intercellular contacts. Absence or reduction of CD18 leads to leukocyte adhesion deficiency (LAD1) and related immune dysfunctions. CD18 is also targeted by Mannheimia haemolytica leukotoxin, mediating cytolysis.

Two transcript variants encoding CD18 have been identified.
