Product Specific Information

The 6.7 monoclonal antibody reacts with human and non-human primate CD18 (beta2 integrin). CD18 is a 90–95 kDa type I transmembrane protein that forms heterodimers with CD11a, CD11b, and CD11c. The CD18/CD11a complex (LFA-1) plays an important role in cellular adhesion and inflammatory reactions.

CD18 is broadly expressed on lymphocytes and monocytes, with lower expression on granulocytes. The 6.7 antibody can induce proliferation of resting T cells cultured with monocytes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human blood cells. Recommended usage: 5 µL (0.6 µg) per test in a final volume of 100 µL (10^5–10^8 cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10^3–10^8 cells).

Target Information

CD18 (integrin beta 2 subunit) is a 90 kDa transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11 molecules (CD11a–d) to create β2 integrins:

• αLβ2 (CD11a/CD18, LFA-1)
• αMβ2 (CD11b/CD18, Mac-1, CR3)
• αXβ2 (CD11c/CD18)
• αDβ2 (CD11d/CD18)

These integrins mediate cell adhesion and signaling, essential for leukocyte migration and intercellular contact.
Deficiency or mutation in CD18 causes leukocyte adhesion deficiency type I (LAD1) and related immune disorders. CD18 also serves as a receptor for Mannheimia haemolytica leukotoxin.
