Product Specific Information

The 6.7 monoclonal antibody reacts with human and non-human primate (rhesus or cynomologus) CD18, also known as beta2 integrin. CD18 is a 90–95 kDa type I transmembrane protein that forms heterodimers with CD11a, CD11b, and CD11c. The CD18/CD11a complex (LFA-1) plays a key role in cellular adhesion and inflammatory responses.

• Expressed broadly by lymphocytes and monocytes; granulocytes express lower levels.
• Reported to induce proliferation of resting T cells cultured with monocytes.
• Pre-titrated and tested for flow cytometric analysis of normal human blood cells.
• Recommended use: 5 µL (0.8 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD18 (integrin beta 2 subunit) is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11a–d to create β2 integrins:

• αLβ2 (CD11a/CD18, LFA-1)
• αMβ2 (CD11b/CD18, Mac-1, CR3)
• αXβ2 (CD11c/CD18)
• αDβ2 (CD11d/CD18)

These integrins mediate leukocyte adhesion and migration.
Defects in CD18 cause leukocyte adhesion deficiency type I (LAD1) and may contribute to psoriasiform skin disease.
CD18 also serves as a receptor for Mannheimia haemolytica leukotoxin, mediating cytolysis.
