Product Specific Information

The 6.7 monoclonal antibody reacts with human and non-human primate (rhesus or cynomolgus) CD18, also known as beta2 integrin.
CD18 is a 90–95 kDa type I transmembrane protein that forms heterodimers with CD11a, CD11b, and CD11c. The CD18/CD11a complex, known as LFA-1, is involved in cellular adhesion and inflammatory responses.

CD18 is expressed broadly by human lymphocytes and monocytes, with lower expression in granulocytes. The 6.7 antibody can induce proliferation of resting T cells cultured with monocytes.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and tested by flow cytometry of normal human blood cells
• Recommended Use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample to antibody cocktails or master mixes before combining with cells.
• For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD18 (integrin beta 2 subunit) is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11a–d to create leukocyte (β2) integrins:

• αLβ2 (CD11a/CD18, LFA-1)
• αMβ2 (CD11b/CD18, Mac-1, CR3)
• αXβ2 (CD11c/CD18)
• αDβ2 (CD11d/CD18)

These integrins mediate cell adhesion and signaling, essential for leukocyte migration and intercellular contact.
Deficiency of CD18 leads to leukocyte adhesion deficiency type I (LAD1).
CD18 also serves as a receptor for Mannheimia haemolytica leukotoxin, mediating cytolysis.

Two transcript variants encoding CD18 have been identified.
