Product Specific Information

The 6.7 monoclonal antibody reacts with human and non-human primate (rhesus or cynomolgus) CD18, also known as beta2 integrin.
CD18 is a 90–95 kDa type I transmembrane protein that heterodimerizes with the α chain of CD11a, CD11b, and CD11c.
The CD18/CD11a complex, also known as LFA-1, plays a critical role in cellular adhesion and inflammatory responses.

• Expressed broadly by human lymphocytes and monocytes
• Granulocytes express CD18 at lower levels
• Reported to induce proliferation of resting T cells cultured with monocytes

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry on normal human blood cells
Recommended use: 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation over the previous CellBlox.
Add 5 µL of CellBlox Plus Blocking Buffer per sample for optimal staining.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing nucleic acid dye structures that provide engineered fluorescent signatures with reduced spillover and spread.

Target Information

CD18 (integrin beta 2 subunit) is a 90 kDa type I transmembrane protein expressed on all leukocytes.
It forms heterodimers with CD11 molecules to create leukocyte (β2) integrins:

• αLβ2 (CD11a/CD18, LFA-1)
• αMβ2 (CD11b/CD18, Mac-1, CR3)
• αXβ2 (CD11c/CD18)
• αDβ2 (CD11d/CD18)

These integrins are crucial for leukocyte migration and intercellular adhesion.
Defects in CD18 cause leukocyte adhesion deficiency type I (LAD1).
Reduced CD18 expression is associated with psoriasiform skin disease.
CD18 also mediates leukotoxin-induced cytolysis by Mannheimia haemolytica.
