Product Specific Information

1 µg/mL of MA5-45718 was sufficient for detection of TCP1 alpha in 20 µg of 3T3 cell lysate by colorimetric immunoblot analysis using Goat anti-rat IgG:HRP as the secondary antibody. Detects approximately 60 kDa and 92 kDa bands. Cross-reactivity with human HSP60 observed in immunoblot. Weak reactivity with Saccharomyces cerevisiae due to epitope sequence variation (AKLRS vs AKLRA). In C. elegans, reacts with TCP1 alpha and another CCT subunit. In plants, recognizes TCP1 of Pisum sativum and Arabidopsis thaliana TCP1 over the epitope region AKLRA. Also reacts with a subunit of a specialized chaperonin involved in phytochrome folding.

Target Information

T Complex Polypeptide 1 (TCP-1) is a ~60 kDa protein constitutively expressed in most eukaryotic cells and upregulated during spermatogenesis. It is a cytosolic subunit of a hetero-oligomeric chaperone complex involved in folding actin and tubulin. Chaperonins recognize and stabilize folding intermediates, sharing features with HSP70 such as high abundance, stress induction, and ATPase activity. The chaperonin family includes mitochondrial HSP60, E. coli GroEL, plastid Rubisco-subunit binding protein, and archaeal TF55. TCP-1 shares ~40% identity with TF55 but minimal similarity to HSP60 and GroEL.
