1 µg/mL of MA5-45720 was sufficient for detection of TCP1 alpha in 20 µg of 3T3 cell lysate by colorimetric immunoblot analysis using Goat anti-rat IgG:HRP as the secondary antibody. Detects approximately 60 kDa and 92 kDa bands. Cross-reactivity with human HSP60 has been observed in immunoblot analysis. Reacts weakly with Saccharomyces cerevisiae, consistent with the epitope sequence AKLRS (instead of AKLRA). In C. elegans, it reacts with TCP1 alpha and another CCT subunit. In plants, it recognizes TCP1 of Pisum sativum and the Arabidopsis thaliana TCP1 sequence over the region of the epitope AKLRA. It also reacts with a subunit of a specialized chaperonin involved in phytochrome folding.

Target Information

The T Complex Polypeptide 1 (TCP-1) is an approximately 60 kDa protein constitutively expressed in almost all eukaryotic cells and upregulated during spermatogenesis. It is found in the cytosol as a subunit of a hetero-oligomeric chaperone complex that assists in folding of actin and tubulin. Chaperonins recognize and stabilize polypeptide intermediates during folding, assembly, and disassembly, sharing characteristics with Heat Shock Protein 70 (HSP70), including high abundance, stress induction, and ATPase activity. The chaperonin family includes mitochondrial HSP60, E. coli GroEL, plastid Rubisco-subunit binding protein, and archaeal TF55. TCP-1 shares ~40% identity with TF55 but minimal similarity to HSP60 and GroEL.
