Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IFN-gamma. Anti-Human IFN-gamma-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect Human IFN-gamma by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human IFN-gamma (500-P32) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Human IFN-gamma.

Western Blot:
To detect Human IFN-gamma by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human IFN-gamma is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

IFN gamma (Interferon gamma, Type II interferon)
IFN gamma is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and cytokines. It is a dimeric glycoprotein consisting of two 146 amino acid subunits (approx. 45 kDa as a homodimer). On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

Functions include:

• Antiviral and tumor antiproliferative activity
• Induction of class I and II MHC
• Macrophage activation
• Enhanced immunoglobulin secretion by B lymphocytes
• Cytokine regulation and synergistic action with other cytokines

Activation occurs through IFN gamma receptor I and II, triggering the JAK-STAT pathway. Human IFN gamma shows ~40% sequence homology with mouse IFN gamma and does not cross-react with mouse.

Regulation:
Upregulated by IL-2, FGF basic, and EGF; downregulated by vitamin D3 or DMN.
Mutations in the IFN gamma gene are associated with aplastic anemia.
