Preparation
Produced from sera of rabbits immunized with highly pure Recombinant Human IFN-gamma. The anti-Human IFN-gamma-specific antibody was purified by affinity chromatography and subsequently biotinylated.

Sandwich ELISA
To detect Human IFN-gamma by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human IFN-gamma (500-P32) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human IFN-gamma.

Western Blot
To detect Human IFN-gamma by Western blot, use at 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human IFN-gamma is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and other cytokines. It is a dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits. IFN gamma exhibits species-specific biological activity and does not cross-react with mouse IFN gamma.
Functions include antiviral activity, tumor antiproliferative effects, induction of MHC class I and II, macrophage activation, and enhancement of immunoglobulin secretion by B cells. It acts via the JAK-STAT pathway through IFN gamma receptor I and II.
Human IFN gamma shares ~40% sequence homology with the mouse counterpart and is regulated by IL-2, FGF basic, and EGF, while downregulated by vitamin D3 or DMN. Mutations in the IFN gamma gene are associated with aplastic anemia.
