Product Specific Information
- Tested in direct ELISA (use antibody at 2–4 µg/mL for coating).
- For immunohistochemistry (PFA-fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
B7-H4 (B7x, VTCN1) is a costimulatory protein functioning as a negative regulator of T-cell mediated immunity.
Although its receptor is unknown, B7-H4 is believed to deliver an inhibitory signal to T-cells, suppressing proliferation, cell cycle progression, and interleukin-2 production.
B7-H4 deficient mice show minimal immune alteration, suggesting its role in fine-tuning immune responses.
It is expressed on activated T-cells, B-cells, monocytes, and dendritic cells. Aberrant expression is linked to cancers of the lung, breast, and ovary.
B7-H4 is primarily located on lymphoid cell membranes and acts as an immunoinhibitory protein interacting with T lymphocyte receptors. Overexpression may aid tumor cells in evading immune detection, particularly in ovarian and breast cancers.
VTCN1 may serve as a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
