Product Specific Information

  • For immunohistochemistry (PFA-fixed samples): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

B7-H4 (also known as B7x, VTCN1) is a co-stimulatory protein functioning as a negative regulator of T-cell mediated immunity. It delivers inhibitory signals to T-cells, preventing proliferation, cell cycle progression, and interleukin-2 production.
B7-H4 is expressed on activated T-cells, B-cells, monocytes, and dendritic cells. Aberrant expression has been linked to cancers of the lung, breast, and ovary.
Overexpression may allow tumor cells to suppress T-cell immunity and promote tumor progression. VTCN1 is also considered a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
