Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues: Heat sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
B7-H4 (also known as B7x, VTCN1) is a costimulatory protein functioning as a negative regulator of T-cell mediated immunity. Although its receptor is unknown, B7-H4 delivers inhibitory signals to T-cells, suppressing proliferation, cell cycle progression, and interleukin-2 production.
It is expressed on activated T-cells, B-cells, monocytes, and dendritic cells. Aberrant expression has been linked to cancers of the lung, breast, and ovary. Overexpression may enable tumor cells to evade immune surveillance. VTCN1 is also considered a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
