Product Specific Information

This antibody is specific for endogenous levels of the ~100 kDa CtIP protein phosphorylated at Ser327. Immunolabeling is completely eliminated by treatment with lambda-phosphatase.


Target Information

CtIP is a 125 kDa protein originally found to interact with transcription repressor CtBP through the PLDLS motif (CtBP-Interacting Protein), suggesting a role in transcription regulation. It also interacts with BRCA1 via the C-terminal BRCT domains, indicating a potential tumor suppressor function. The CtIP–BRCA1 interaction can be disrupted by DNA-damaging agents such as UV or gamma irradiation.
Li et al. (Nature 406, 210–215, 2000) demonstrated that ATM phosphorylates CtIP at serine residues 664 and 745; mutation of these sites prevents BRCA1 dissociation from CtIP, leading to persistent repression of BRCA1-dependent induction of GADD45 upon ionizing radiation.
