Product Specific Information
This antibody is specific for endogenous levels of the ~345 and 395 kDa Ki-67 isoforms. Additional bands are thought to be breakdown products of these two isoforms.
Target Information
Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly during late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and the nucleolus.
Ki-67 shows a cell cycle-dependent localization pattern:
- G1 phase: Perinucleolar expression
- G2 phase: Expression in the nuclear matrix
- M phase: Expression on chromosomes
As Ki-67 is absent in G0 cells and increases steadily from G1 through mitosis, it serves as a reliable marker of cell proliferation. Ki-67 antibodies are widely used to determine the growth fraction in neoplasms, with strong correlation between low Ki-67 index and histologically low-grade tumors. It is also routinely used as a neuronal marker for cell cycling and proliferation studies.
