Store vial at 4°C prior to restoration. For extended storage, aliquot contents and freeze at -20°C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

Anti-EBI-3 Antibody / Anti-IL27 is an IgG fraction antibody purified from monospecific antiserum by a multi-step process including delipidation, salt fractionation, and ion-exchange chromatography followed by extensive dialysis. This antibody is specific for human EBI3 protein. A BLAST analysis suggests cross-reactivity with EBI3 from human sources (100% homology) and possible cross-reactivity with chimpanzee (97% homology). Cross-reactivity with other species has not been determined.

Target Information

EBI3 (Epstein-Barr virus Induced-3) is a secreted glycoprotein of the hematopoietin receptor family. It plays a regulatory role in Th2-type immune responses and Th2-mediated tissue inflammation, possibly via control of iNKT cell function. EBI3 forms heterodimers with p28 and p35 subunits of IL-12 to generate IL-27 and IL-35, respectively. IL-27, an early product of activated antigen-presenting cells, negatively regulates Th17 differentiation. EBI3 expression is widely distributed and transcriptionally regulated by TLR signaling through MyD88 and NF-κB. It inhibits delayed-type hypersensitivity by suppressing IL-17 and inducing IL-10 production. EBI3 may also modulate tumor metastasis via lung CD8+ T cells, and its deficiency is linked to reduced Th2 cytokine production related to allergic airway inflammation in asthma. Targeted deletion of EBI3 protects mice from lung metastasis.
