Store vial at 4°C prior to restoration. For extended storage, aliquot contents and freeze at -20°C or below. Avoid freeze/thaw cycles. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

This is an IgG preparation of whole rabbit serum purified by DEAE fractionation. The antibody is primarily directed against mature, 17 kDa human IL-1β and is useful for detecting IL-1β in various assays. It also detects primate IL-1β using similar dilutions but does not recognize human IL-1α.

In ELISA and other immunoassays, the antibody recognizes about 10% of the non-denatured 31 kDa IL-1β precursor but primarily detects the 17 kDa mature form. In immunoblots, heating samples in SDS (with or without reducing agents) denatures the 31 kDa precursor, allowing detection as a 35 kDa band due to unfolding. The antibody detects both the 17 kDa mature band and the 35 kDa precursor band in cell lysates and supernatants.

Target Information

Interleukin-1 beta (IL-1β) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is synthesized as a 31 kDa inactive pro-form that accumulates in the cytosol. Cleavage into the active 17 kDa form requires inflammasome activation and caspase-1 processing. IL-1β lacks a signal sequence for classical secretion and is released via an alternate pathway.

IL-1β signals through IL-1RI and IL-1RII receptors, shared with IL-1α. Its activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which competes for receptor binding. IL-1β is central to innate immune responses, inducing proinflammatory cytokines and acute-phase reactions. Elevated IL-1β levels are associated with chronic inflammatory diseases, making IL-1β neutralizing antibodies of therapeutic interest.
