This antibody reacts with an extracellular epitope of CD45RA, a 205–220 kDa single-chain type I glycoprotein, which is a variant of CD45 (CD45RA isoform). CD45RA is expressed on most B lymphocytes, resting and naïve T lymphocytes, medullary thymocytes, and monocytes.
Target Information
CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising about 10% of all surface proteins in lymphocytes. It plays a critical role in lymphocyte development and antigen signaling by regulating Src-family kinases.
CD45 exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. Isoform expression depends on cell type and physiological state. CD45 also contributes to integrin-mediated signaling, cell survival, and apoptosis regulation. It is expressed in hematopoietic cells except mature red cells and absent in other tissues. CD45 dysfunction is associated with severe combined immunodeficiency and Hepatitis C.
