Target Information
SIK3 (QSK) is a serine/threonine-protein kinase belonging to the QIK subfamily. Phosphorylation by LKB1 via 14-3-3 binding enhances catalytic activity and localizes SIK3 to punctate cytoplasmic structures. Overexpression promotes G1/S cell cycle progression, notably in ovarian cancer. Mutations (H331L, A1103V) occur frequently in breast cancer. SIK3 acts as a tumor-associated antigen and positive regulator of mTOR signaling through DEPTOR degradation. It plays a role in mTOR signaling regulation during chondrocyte differentiation and negatively regulates cAMP signaling via CRTC2/TORC2 and CRTC3/TORC3.
