• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, playing a key role in signal transduction and T cell differentiation and activation.
CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC class I molecules.
It is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
Binding of MHC-I/peptide complexes on antigen-presenting cells triggers Lck activation, leading to cytokine production, motility, and cytotoxic T lymphocyte activation.
Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
