Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family that plays a key role in signal transduction, T cell differentiation, and activation. CD8 is primarily expressed on cytotoxic T cells as a disulfide-linked heterodimer (CD8α and CD8β) and functions as a co-receptor with the T cell receptor (TCR) for MHC class I molecules.
CD8α is essential for MHC-I binding and is also expressed as a homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells. Interaction with MHC-I/peptide complexes on antigen-presenting cells recruits Lck kinase, leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
