Product Specific Information

Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells. IL-7 binding triggers signal transduction via tyrosine kinase pathways including Jak/STAT.

CD127 expression is down-regulated on CD4+CD25+ regulatory T cells (Tregs). CD4+CD25+CD127^low cells are highly enriched for regulatory T populations expressing Foxp3 and showing suppressive activity.

Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7.

Each product contains:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for use at 5 µL (0.2 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Product # C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Use 5 µL per sample (per 100 µL cell suspension)
• Add to antibody cocktails before mixing with cells

Target Information

CD127 (Interleukin-7 receptor α) is a glycoprotein regulating lymphopoiesis. It forms an α/γ heterodimer receptor complex (IL7R) that mediates IL-7-dependent signaling. The γ chain activates transduction, while the α chain provides specificity. CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
Defects in CD127 are associated with severe combined immunodeficiency (SCID).
