Product Specific Information
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha).
CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R), a type I glycoprotein (~75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells.
IL-7 binding triggers signal transduction through tyrosine kinase pathways such as Jak/STAT. IL-7 is essential for lymphocyte development and homeostatic T-cell proliferation. CD127 expression is down-regulated on CD4+CD25+ regulatory T cells, allowing improved identification of suppressive T reg populations.
Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Applications Reported: Flow cytometric analysis
Applications Tested: 5 µL (0.2 µg)/test; suitable for 10⁵–10⁸ cells/sample.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
- CellBlox Plus Blocking Buffer (Product #C001T06F01) is required for optimal staining.
- Add 5 µL per sample or per 100 µL cell sample (10³–10⁸ cells).
- NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures with minimized spillover.

Target Information
CD127 (Interleukin-7 receptor, IL7R) regulates lymphopoiesis via an alpha/gamma chain heterodimer.
The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling.
CD127 promotes proliferation of precursor B cells, thymocytes, and mature CD4+/CD8+ T cells.
Dysfunction is associated with severe combined immunodeficiency (SCID).
