Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-17A. Anti-Human IL-17A-specific antibody was purified by affinity chromatography using an immobilized Human IL-17 matrix.
Sandwich ELISA:
To detect Human IL-17A by sandwich ELISA (100 µL/well), use 0.5–2.0 µg/mL of this antibody. In conjunction with PeproTech Biotinylated Anti-Human IL-17A (500-P07Bt) as detection antibody, it enables detection of at least 0.2–0.4 ng/well of Recombinant Human IL-17A.
Western Blot:
For detection of Human IL-17A by Western Blot, use 0.1–0.2 µg/mL. With compatible secondary reagents, the detection limit for Recombinant Human IL-17A is 1.5–3.0 ng/lane under reducing or non-reducing conditions.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in conditions such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
IL-17A is a 32 kDa disulfide-linked homodimer (136 amino acids) belonging to the IL-17 family (IL-17A–F) and signals via the IL-17 receptor (IL-17R/CDw217).
It induces expression of ICAM-1 in fibroblasts and stimulates epithelial, endothelial, and fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2. IL-17A also supports proliferation and differentiation of CD34+ hematopoietic progenitors into neutrophils. Mouse, rat, and human IL-17A can induce IL-6 secretion in mouse stromal cells, indicating cross-species receptor recognition.
IL-17A regulates NF-κB and MAPK pathways, enhances IL-6 and COX-2 expression, and promotes nitric oxide production.
