Target Information
TRIP12 is an E3 ubiquitin-protein ligase involved in the ubiquitin fusion degradation (UFD) pathway and regulation of DNA repair.
It mediates ubiquitination and degradation of isoform p19ARF/ARF of CDKN2A, a tumor suppressor required for p53/TP53 activation under oncogenic stress.
In cancer cells, TRIP12 and p19ARF/ARF are located in different compartments, preventing degradation.
TRIP12 also catalyzes ubiquitination of NAE1 and SMARCE1, leading to their degradation.
Acts as a key regulator of DNA damage response by suppressing RNF168, preventing excessive accumulation of ubiquitinated chromatin at damaged chromosomes.
