Product Specific Information

The monoclonal antibody 42D1 recognizes the CD85d protein (ILT4, LIR-2), a member of the ILT/LIR/MIR receptor family. CD85d is a 110 kDa single transmembrane receptor with three Ig-like domains and a cytoplasmic domain containing ITIMs. Expression is confined to monocytes, macrophages, and dendritic cells. Its ligand includes non-classical class I HLA-G, -B, and G1. Upon ligand binding, SHP-1 interacts with ITIMs, contributing to immune tolerance during fetal implantation and possibly organ allograft rejection.

Crosslinking of CD85d with the 42D1 antibody inhibits calcium flux and enhances HLA-G tetramer staining, restoring the stimulating activity of tolerogenic dendritic cells.

Applications Reported: Flow cytometric analysis and functional assays.
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimal performance requires titration.

Target Information

The LILRB2 (ILT4) gene is part of the leukocyte immunoglobulin-like receptor (LIR) family located at chromosomal region 19q13.4. The encoded protein belongs to the subfamily B class of LIR receptors with two to four extracellular Ig domains, a transmembrane region, and ITIM motifs. Expressed on immune cells, it binds MHC class I molecules on antigen-presenting cells to inhibit immune stimulation, controlling inflammation and limiting autoreactivity. Multiple transcript variants exist.
