The monoclonal antibody 27D6 recognizes the CD85d (ILT4, LIR-2) protein, a member of the ILT/LIR/MIR receptor family.
CD85d is a 110 kDa transmembrane receptor with 3 Ig-like domains and a long cytoplasmic domain containing ITIM motifs.
Expression is restricted to monocytes, macrophages, and dendritic cells. Its ligand includes non-classical class I HLA-G and -B molecules.
Upon ligand binding, tyrosine phosphatase SHP-1 interacts with ITIM, contributing to immune tolerance during fetal implantation and playing a role in organ allograft rejection.

The 27D6 antibody blocks ligand (HLA-G tetramer) binding and is suitable for in vitro blocking assays and flow cytometric analysis.
For flow cytometry, use ≤1 µg/test (100 µL final volume, cell number 10⁵–10⁸ cells/test).
Careful titration is recommended for optimal assay performance.

Target Information

The LILRB2 (ILT4) gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosomal region 19q13.4.
It encodes a B-class receptor with extracellular Ig domains, a transmembrane domain, and ITIM motifs.
ILT4 is expressed on immune cells, binds MHC class I molecules, and transduces inhibitory signals to regulate immune responses, inflammation, and autoreactivity.
Multiple transcript variants encoding different isoforms exist.
