Target Information
Adrenomedullin (ADM) is a hypotensive peptide belonging to a peptide superfamily including calcitonin gene-related peptide (CGRP) and amylin.
Originally identified in the adrenal medulla, ADM is expressed in most contractile cells and upregulated during sepsis and ischemia.
Three distinct receptors bind ADM:
- ADM receptor (L1)
- RDC-1
- Calcitonin Receptor-Like Receptor (CRLR)
CRLR associates with receptor activity-modifying proteins (RAMPs):
- RAMP1 → CGRP binding
- RAMP2/3 → ADM binding
ADM receptor is a seven-transmembrane G protein-coupled receptor, highly expressed in heart, brain, skeletal muscle, immune system, adrenal gland, and liver. It mediates ADM signaling through G proteins that activate adenylate cyclase.
