Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (gamma and delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to T cell activation and proliferation.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (1 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related immunoglobulin superfamily members encoded on human chromosome 11. These subunits associate with cytoplasmic signal transduction molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Decreased T lymphocyte percentages may occur in autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
