Product Specific Information

The 104 monoclonal antibody reacts with the mouse CD45 molecule, the leukocyte common antigen (LCA) in CD45.2-expressing mouse strains such as C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize dilution for specific assays.
  • Fluorochrome: Brilliant Ultra Violet™ 805 (BUV805), emission at 797 nm, suitable for 355 nm UV laser.
  • Light Sensitivity: Protect from light to prevent photo-induced oxidation.
  • Fixation: Compatible with IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.
  • Polymer Dye Panels: Use Super Bright Complete Staining Buffer (Product #SB-4401) or Brilliant Stain Buffer (Product #00-4409-75) to minimize non-specific interactions.
  • Methanol Compatibility: Short-term methanol fixation is acceptable; avoid long-term methanol exposure (>1 hour).
  • Excitation/Emission: 355 nm / 797 nm; Laser: Ultraviolet.

Target Information

CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells and constitutes about 10% of lymphocyte surface proteins. It plays a key role in lymphocyte development and antigen signaling by regulating Src-family kinases. CD45.2 exists in multiple isoforms due to alternative splicing, differing in extracellular domains. It also participates in integrin-mediated signaling, cell survival, and apoptosis regulation.
