Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling and homing to peripheral lymphoid tissues.

Applications Reported: Flow cytometric analysis.

Applications Tested: Flow cytometric analysis of mouse splenocytes; ≤0.06 µg/test. A test is defined as antibody amount for staining in 100 µL volume. Cell count range: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm, suitable for cytometers with a 355 nm UV laser. Ensure instrument compatibility.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product #SB-4401) or Brilliant Stain Buffer (Product #00-4409) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal impact on brightness. Clone-specific performance should be empirically determined.

Methanol Compatibility:
BUV805 is compatible with short-term methanol fixation (<1 hour). Avoid long-term methanol exposure.

355 nm / 797 nm / Ultraviolet Laser.

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:

• Cytoplasmic region

CD62L is constitutively expressed on leukocytes including lymphocytes, monocytes, and polymorphonuclear cells, except some memory T cells.
