Product Specific Information

The monoclonal antibody 1F3 recognizes human TRAF-1, a multifunctional adapter molecule and member of the TNF receptor-associated factor (TRAF) protein family. TRAF-1 regulates cell survival and apoptosis, mediates NF-κB and JNK signaling, and forms a heterodimer with TRAF-2. It interacts with receptors such as CD30, CD40, CD357 (AITR), and cytoplasmic proteins like NIK, RIP2, A20, and TRIP. Expression can be induced by Epstein-Barr Virus (EBV) and TNF-α.

Applications Reported: Suitable for microscopy, immunohistochemical staining, and immunocytochemistry.
Applications Tested: Validated in IHC on formalin-fixed, paraffin-embedded human tissue using low pH antigen retrieval at ≤5 µg/mL. Optimal titration recommended.

Filter Recommendation: Use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Compatible with standard Alexa Fluor® 555 or TRITC filters.

Target Information

TRAF-1 is a member of the TNF receptor-associated factor (TRAF) family mediating signal transduction from TNFR superfamily receptors. It forms a heterodimer with TRAF2 essential for TNF-α-mediated activation of MAPK8/JNK and NF-κB. The TRAF1–TRAF2 complex interacts with inhibitor-of-apoptosis proteins (IAPs), contributing to anti-apoptotic signaling. Expression is inducible by EBV, and LMP1 interacts with TRAF proteins linking B lymphocyte transformation to TNFR signaling. Three transcript variants encode two isoforms.
