Product Specific Information

The eBioMLC-51 monoclonal antibody recognizes mouse Nanog, a multidomain homeobox transcription factor that maintains pluripotency of embryonic stem cells independent of LIF/Stat3. Nanog is expressed in early embryos, the inner cell mass of the blastocyst, embryonic stem (ES) cells, and embryonic germ (EG) cells. Its expression overlaps but is not identical to Oct4 and is downregulated upon differentiation, making it a marker for undifferentiated stem cells.

Nanog contains activation domains CD2 and WR in the C-terminus and one in the N-terminus. The CD2 domain is unique to Nanog, while the NK2 DNA binding domain is well conserved.

Immunoblotting using eBioMLC-51 reveals a band at ~45 kDa in F9 embryonal carcinoma cell lysate but not in NIH3T3 or mouse spleen lysates.

For intracellular staining of mouse Nanog using fluorochrome-conjugated eBioMLC-51, it is essential to use the eBioscience Foxp3 Staining Buffer Set (Product #00-5523-00).

Target Information

NANOG (Nanog homeobox) is a divergent homeodomain protein involved in maintaining pluripotency and differentiation of embryonic stem cells. NANOG mRNA is expressed in pluripotent mouse and human cells, absent in differentiated cells. Human NANOG shares 52% identity with mouse NANOG (85% in homeodomain).
Human NANOG maps to 12p13.31; mouse NANOG to 6F2. Expression is detected in the inner cell mass of the blastocyst.

High NANOG expression is observed in undifferentiated N-Tera embryonal carcinoma cells. NANOG regulates proliferation and self-renewal of ES cells, playing a role in stem cell creation for regenerative therapies. Induced pluripotent stem (iPS) cells can be generated using NANOG, Sox2, Klf4, and c-Myc expression plasmids, avoiding viral introduction for safer applications. Overexpression promotes S-phase entry and proliferation.
Diseases linked to NANOG dysfunction include teratocarcinoma and germ cell/embryonal cancer.
