Product Specific Information

The 5C6 monoclonal antibody reacts with human NKG2D, a 42 kDa lectin-like molecule expressed by NK cells, γδ T cells, and subsets of CD4⁺ and CD8⁺ T cells. NKG2D forms complexes with DAP10, functioning as a co-stimulatory receptor for NK activation. Its counter-receptors MICA/MICB are expressed on various epithelial tumors (lung, breast, kidney, ovary, prostate, colon).

5C6 and 1D11 clones block binding of soluble MICA to γδ TCR T cell clones and inhibit lysis by these cells, while also inducing redirected lysis through FcR-bearing P815 cells.

For functional assays, use Functional Grade purified 5C6 (Product #16-5879).

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.5 µg per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information

Natural killer (NK) cells mediate lysis of certain tumor and virus-infected cells without prior activation. They regulate humoral and cell-mediated immunity and express several calcium-dependent (C-type) lectins involved in NK function. The NKG2 gene family encodes type II transmembrane proteins with C-type lectin domains, located within the NK complex region rich in NK-specific lectin genes.
