Description:
The MAR-1 monoclonal antibody reacts with the Fc epsilon Receptor I alpha subunit (FcεRIα), an IgE-binding component lacking signal-transducing ability. FcεRIα is expressed on mast and basophil cells and is up-regulated by IgE presence. It forms a tetrameric complex with one beta and two gamma subunits, which contain ITAM motifs. The FcεRI complex is crucial for initiating IgE-mediated allergic reactions.

Applications Reported:
Suitable for flow cytometric analysis, immunoprecipitation, and immunohistology of frozen tissue sections. Also used in mast cell degranulation assays. For functional assays, use Functional Grade purified MAR-1 (Product #16-5898).

Applications Tested:
Validated in flow cytometric analysis of MC/9 mouse mast cell line. Recommended ≤0.25 µg/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best assay performance.

Target Information

FceR1 (FcεRIα) is part of the high-affinity IgE receptor complex consisting of one α, one β, and two γ subunits. The β and γ subunits mediate signal transduction. This complex triggers allergic responses by coupling allergen-bound IgE with mast cells, leading to histamine release and synthesis of prostaglandins and leukotrienes—key mediators of hypersensitivity reactions such as hay fever and asthma.
