Strand-specific Illumina sequencing libraries from picogram RNA inputs of any quality

The SMARTer Stranded Total RNA-Seq Kit v2 - Pico Input Mammalian enables efficient preparation of Illumina sequencing libraries from picogram-level total RNA inputs (250 pg–10 ng).
It supports processing of high-quality, partially degraded, or low-quality RNA, including RNA from FFPE or LCM samples.

This updated version of the original SMARTer kit features:

• Improved sequencing performance across all Illumina platforms
• Streamlined, user-friendly purification workflow
• Strand information retention during library prep
• Complete workflow (library prep + purification) in approximately 6 hours

Ribosomal cDNA depletion technology

Ribosomal RNA (rRNA) can comprise up to 90% of total RNA. Depleting these abundant transcripts reduces sequencing costs and improves mapping statistics.
This kit’s proprietary technology depletes ribosomal cDNA using probes specific to mammalian rRNA and some mitochondrial RNA, ensuring sufficient material remains for high-quality library construction even from very low input amounts.

Enhanced sequencing performance

Libraries generated with this kit:

• Achieve high cluster passing filter (%PF) without PhiX addition
• Perform reliably on two-channel SBS Illumina systems (NextSeq®, MiniSeq™, HiSeq® 3000/4000)
• Yield more unique transcripts, fewer reads mapping to rRNA/mtRNA, and lower duplicate rates
• Include a new PCR buffer formulation for improved purification efficiency

Kit formats and compatibility

Available in 12-, 48-, 96-, and 192-reaction sizes:

• High-throughput versions (Cat. Nos. 634413 & 634414) include 12 reverse and 8 forward primers for up to 96 index combinations.
• Versions without indexing primers (Cat. Nos. 634418 & 634419) are compatible with SMARTer RNA Unique Dual Index Kits (Cat. Nos. 634451, 634452 & 634457, sold separately).

The original SMARTer Stranded Total RNA-Seq Kit - Pico Input Mammalian remains available and supported.
