Product Specific Information

The RPA-T8 monoclonal antibody recognizes the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8α/β heterodimer or CD8α/α homodimer. It is found on most thymocytes and subsets of mature T cells and NK cells. CD8 binds MHC class I and associates with p56lck kinase, contributing to T cell development and activation.

• Applications Tested: Pre-titrated for flow cytometry on normal human peripheral blood cells
• Recommended Usage: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Super Bright 600 Dye:

• Excitation: 405 nm (Violet laser)
• Emission: 600 nm (recommend 610/20 bandpass filter)
• Sensitive to light; protect from photo-oxidation
• Use Super Bright Complete Staining Buffer (SB-4401) when multiple Super Bright dyes are used to minimize non-specific interactions

Fixation and Storage:

• Samples can be stored up to 3 days at 2–8°C in the dark using
  • IC Fixation Buffer (00-8222), or
  • 1-step Fix/Lyse Solution (00-5333)
• Clone-specific performance after fixation should be empirically determined.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family involved in T cell signal transduction and activation. CD8 is mainly expressed on cytotoxic T cells as a CD8α/β heterodimer, acting as a co-receptor for MHC class I molecules. CD8α is critical for MHC-I binding and is also found on subsets of NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes triggers Lck kinase recruitment, leading to activation of cytotoxic T lymphocytes (CTLs) and immune responses against pathogens and tumors. Differentiation of naive CD8⁺ T cells into CTLs is promoted by IL-2, IL-12, and TGF-β1.
