Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8a is found on most thymocytes and subsets of mature T cells and NK cells. It binds MHC class I molecules and, via association with p56lck, plays a critical role in T-cell development and activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry on normal human peripheral blood cells at 5 µL (0.125 µg) per test. A test corresponds to staining in 100 µL final volume, with 10⁵–10⁸ cells/test depending on assay setup.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection with a 610/20 bandpass filter. Ensure instrument compatibility.

Usage Recommendations:

• For panels using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
• Protect from light due to photo-induced oxidation sensitivity.
• Fixed samples can be stored up to 3 days at 4°C in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor for MHC class I molecules, aiding in T cell receptor (TCR) signaling, differentiation, and activation. CD8 is expressed as CD8αβ heterodimers on cytotoxic T cells and as CD8αα homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Binding of MHC-I/peptide complexes triggers Lck kinase recruitment, leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
