Target Information
Metadherin (Metastasis adhesion protein), also known as MTDH or LYRIC, is a novel protein that localizes with tight junction proteins ZO-1 and occludin in polarized epithelial cells. It is recruited during maturation of the tight junction complex, rather than acting as a structural component. Metadherin is overexpressed in breast cancer tissue and tumor xenografts, while lower levels are found in normal breast tissue. It binds to lung vasculature — a common site of breast cancer metastasis — through a C-terminal extracellular domain segment. Blocking this domain with antibodies or inhibiting metadherin using siRNA has been reported to inhibit breast cancer metastasis.
