Product Specific Information

Description:
The SK1 monoclonal antibody reacts with the human CD8a molecule, an approximately 32–34 kDa cell surface receptor expressed as a heterodimer with CD8 beta (CD8α/β) or as a homodimer (CD8α/α). CD8a is expressed on most thymocytes and subsets of mature T cells and NK cells. CD8 binds to MHC class I and, via association with protein tyrosine kinase p56lck, participates in T-cell development and activation.

Applications Reported:
This SK1 (SK-1) antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry on normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test (in 100 µL final volume). Cell numbers may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm.
Recommended filter: 610/20 bandpass.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal loss of brightness.

Target Information

CD8 (Cluster of Differentiation 8):
A type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction, T-cell differentiation, and activation.
CD8 is expressed on T cells as CD8α/β heterodimers or CD8α/α homodimers, functioning as a co-receptor with TCR for MHC-I molecules.
It is also found on subsets of T cells, NK cells, monocytes, and dendritic cells.
Binding of MHC-I/peptide complexes triggers recruitment of Lck kinase, leading to cytokine production and cytotoxic T lymphocyte (CTL) activation.
Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
