Product Specific Information

Description:
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8 binds to MHC class I and, through association with tyrosine kinase p56lck, plays a role in T cell development and activation.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Use ≤0.5 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize concentration empirically.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by a 405 nm violet laser and emits at 600 nm. Recommended detection with a 610/20 bandpass filter.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark.

405 nm / 600 nm (Violet Laser)

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction, T cell differentiation, and activation.
CD8 is primarily expressed on T cells as CD8αβ heterodimers, functioning as a co-receptor with TCR for MHC-I molecules.
A subset of T cells, NK cells, monocytes, and dendritic cells express CD8αα homodimers.
Engagement of MHC-I/peptide complexes recruits Lck kinase, leading to lymphokine production and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are crucial for pathogen and tumor cell clearance, with differentiation enhanced by IL-2, IL-12, and TGF-β1.
