Product Specific Information

The eBioHP40 antibody reacts with the p40 subunit of human IL-12 p70 and IL-23.
IL-12 is a 70 kDa heterodimeric cytokine composed of p40 and p35 subunits, secreted mainly by activated monocytes, macrophages, and dendritic cells.
IL-23 is a related heterodimeric cytokine composed of p40 and p19 subunits.

The eBioHP40 antibody shows lower background staining than C8.6 and recognizes a different epitope.
It is validated for intracellular staining followed by flow cytometric analysis and pre-titrated for use at 5 µL (0.03 µg) per test.
A test is defined as staining a cell sample in a final volume of 100 µL (typically 10⁵–10⁸ cells/test).

eFluor® 660 replaces Alexa Fluor® 647, emits at 659 nm, and is excited with a red laser (633 nm).
Ensure instrument compatibility with this fluorochrome.

Target Information

IL-12 is produced by dendritic cells, neutrophils, macrophages, and lymphoblastoid cells upon antigenic stimulation.
It is encoded by IL-12A (p35) and IL-12B (p40).
IL-23 shares the IL-12B subunit and includes IL-23A (p19).

Functionally, IL-12 increases endothelial adhesion molecule-1 expression, directs leukocytes toward tumors, and promotes ischemic-hemorrhagic necrosis of tumor tissue.
IL-23 stimulates memory T-cells and is essential for Th17 cell activation and maintenance, contributing to neutrophil chemokine and pro-inflammatory cytokine upregulation.
