Product Specific Information

The DB-1 monoclonal antibody reacts with rat interferon gamma (IFN-γ), a 15–20 kDa cytokine secreted by Th1 cells.

  • Applications Reported: Intracellular staining followed by flow cytometric analysis.
  • Applications Tested: Tested for intracellular staining and flow cytometric analysis of stimulated rat splenocytes.
  • Recommended Use: ≤0.5 µg per test (in 100 µL final volume; 10⁵–10⁸ cells/test).
  • Optimization: Antibody should be titrated for best performance.
  • Fluorochrome: eFluor® 660 (replacement for Alexa Fluor® 647).
    • Emission: 659 nm
    • Excitation: 633 nm (Red laser)
    • Ensure instrument compatibility.
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

IFN gamma (Interferon gamma, Type II interferon)
A macrophage activation factor produced primarily by T-lymphocytes and NK cells in response to antigens, mitogens, and cytokines.

  • Dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits
  • Appears as 25, 20, and minor 15.5 kDa bands on SDS-PAGE due to differential glycosylation
  • Highly species-specific biological activity (no cross-reactivity between human and mouse IFN-γ)
  • Functions: antiviral activity, tumor antiproliferative activity, MHC induction, macrophage activation, enhanced B cell immunoglobulin secretion
  • Acts via IFN-γ receptor I/II and JAK-STAT pathway
  • Upregulated by IL-2, FGF-basic, EGF; downregulated by vitamin D3 or DMN
  • Gene mutations associated with aplastic anemia
