Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, T helper cells, and monocytes. CD4 serves as a receptor for HIV. The SK3 clone blocks HIV binding and mixed lymphocyte reaction, recognizing distinct epitopes from RPA-T4.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for use at 5 µL (0.1 µg) per test.
A test corresponds to staining a cell sample (10⁵–10⁸ cells) in 100 µL volume.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer:
Contains improved formulation for optimal staining.

• Add 5 µL per sample to antibody cocktail.
• For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor Technology:
Based on Phiton technology utilizing nucleic acid dye structures engineered for reduced spillover and improved spectral performance.

Target Information

CD4 antigen is a co-receptor for HIV and recognizes MHC class II molecules.
Expressed on T helper cells, monocytes, macrophages, and dendritic cells.
Detected in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut.
Functions in early T-cell activation through association with TCR complex and Lck kinase.
May mediate neuronal damage in infectious and immune-mediated CNS diseases.
Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
