Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor Dyes Compatibility:
Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer Usage:
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample to antibody cocktails before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology:
Based on Phiton technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures and minimized spillover/spread.

Target Information

The CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. Expressed mainly on T helper cells, monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 facilitates T-cell activation and may mediate neuronal damage in infectious and immune-mediated CNS diseases. Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
