Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV; SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes prior to combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD4 antigen is involved in MHC class II recognition and acts as a co-receptor for HIV. It is expressed on T helper cells and other immune cells such as monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 aids T-cell activation via Lck association and may mediate neuronal damage in immune-related CNS diseases.

Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
