Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed on most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and SK3 can block HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

This SK3 (SK-3) antibody is validated for flow cytometric analysis.

Applications Tested

Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.1 µg) per test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test (empirically determined).

Handling Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD4 antigen participates in MHC class II recognition and serves as a co-receptor for HIV. It is mainly expressed in T helper lymphocytes and also found on monocytes, macrophages, and dendritic cells.
Tissue expression includes thymus, lymph nodes, tonsils, spleen, and various non-lymphoid tissues.
CD4 is crucial for early T-cell activation via association with the T-cell receptor complex and Lck kinase, and may contribute to neuronal damage in immune-mediated CNS diseases.
Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
